WRN, or Werner syndrome protein, is a multifunctional DNA deconjugating enzyme that is a member of the RecQ family of deconjugating enzymes.WRN functions in the nucleus with ATP-dependent DNA deconjugating enzyme activity and exonuclease activity from the 3‘ end to the 5’ end. Its mechanism of action involves DNA replication, recombination, damage repair, and telomere maintenance.WRN regulates the cellular response to DNA damage through its involvement in cell cycle regulation and DNA repair signalling pathways, such as interactions with proteins such as p53, RAD50, RAD51, etc.The biological significance of WRN lies in the maintenance of genome integrity and stability, and in the prevention of premature aging and certain types of cancer. Mutations in the WRN gene have been associated with Werner syndrome, a progeroid disease in which patients experience an accelerated aging process and a higher risk of cancer.
