ADAM9, full name A Disintegrin And Metalloproteinase Domain 9, is also known as MDC9 or Meltrin gamma. As a transmembrane metalloproteinase, it combines disintegrin and metalloprotease domains to mediate cell-cell/matrix adhesion and cleave substrates like pro-HB-EGF and pro-MMPs, regulating EGFR and PI3K/Akt signaling pathways.
Overexpression of ADAM9 correlates with non-small cell lung cancer, colorectal cancer, and breast cancer, promoting tumor invasion, metastasis, and angiogenesis. Preclinical studies are exploring ADAM9-targeted therapies—including monoclonal antibodies and small-molecule inhibitors—to block its proteolytic activity and suppress tumor progression.
