BCL11B, or B-cell lymphoma/leukemia 11B gene, also known as CTIP2 or RIT1, plays a crucial role in T cell development and identity maintenance. Initially identified as a potential suppressor of radiation-induced T cell lymphomas, BCL11B's mechanism of action involves inhibiting the NuRD complex and promoting the interaction between β-catenin and TCF4 to activate Wnt signaling, with its expression being directly repressed by BMP signaling. BCL11B functions as a haploinsufficient tumor suppressor in T cell acute lymphoblastic leukemia (T-ALL), working in conjunction with other oncogenic alterations. BCL11B is also associated with neurodevelopment, with its expression being downregulated during oligodendrocyte differentiation, yet it plays a regulatory role in neuronal differentiation and the development of the central nervous system. Abnormal expression of BCL11B is linked to various diseases, including T-ALL and certain neurodevelopmental disorders.
