BFAR, also known as BAR or RNF47, is a multifunctional apoptosis regulator. BFAR is a multi-domain E3 ubiquitin ligase that participates in the negative regulation of IRE1-mediated unfolded protein response, the ubiquitin-dependent proteolysis process mediated by the proteasome, and protein ubiquitination through its binding to caspases, protein-macroprotein bridging activity, and ubiquitin protein ligase activity. BFAR plays a role in the negative regulation of apoptosis and is located in the endoplasmic reticulum and membrane. Abnormal expression of BFAR is associated with various diseases, including tumors, where high expression in CD8+ T cells derived from elderly individuals can inhibit TRM cell production, affecting the bodys anti-tumor immune response.
