CD180, formerly known as RP105, was originally discovered on human B cells and is a TLR homolog. It has also been found on monocytes and some dendritic cells. Mice homozygous for a null mutation in the CD180 gene were impaired in lipopolysaccharide (LPS)-induced B cell proliferation and antibody immunity, suggesting the importance of CD180 in B cell activation. The extracellular MD-1 molecule associated with CD180 is important for its expression on B cells, B cell response to LPS in mice, and activation of human B cells. Anti-CD180 monoclonal antibody (mAb) induces human and mouse B cell proliferation and upregulation of major histocompatibility complex (MHC) class II, CD40 and CD80/CD86. Involved in the recognition of bacterial LPS by TLR-4 and thus involved not only dendritic cells but also innate immunity of B cells.
