CHMP7

Charged Multivesicular Body Protein 7 (CHMP7) is a core component of the endosomal sorting complex required for transport-III (ESCRT-III), mediating membrane scission in processes like endosome maturation, autophagosome closure, and nuclear envelope repair. It interacts with ESCRT-I/II to initiate ESCRT-III assembly, driving membrane remodeling. Common aliases include CHMP7_HUMAN and KIAA0174.
Mutations in CHMP7 are associated with Charcot-Marie-Tooth disease type 2 (CMT2), a peripheral neuropathy causing muscle weakness. Preclinical research explores small molecules targeting CHMP7’s ESCRT interactions to modulate autophagy or nuclear envelope repair, with potential uses in neurodegeneration and cancer—though no clinical drugs are approved yet.
