COMMD9, or COMM domain-containing protein 9, is a conserved member of the COMM family. It forms the CCC complex with CCDC22 and CCDC93, regulating endosomal recycling of membrane proteins and suppressing NF-κB signaling by promoting IκBα degradation. It has no widely recognized aliases beyond its official name.
Dysregulated COMMD9 may contribute to inflammatory disorders via NF-κB dysbalance. Altered expression links to certain cancers where hyperactive NF-κB drives tumor progression. Direct COMMD9-targeting drugs are unavailable, but NF-κB pathway inhibitors (e.g., IKK inhibitors) could indirectly mitigate its pathological roles in disease contexts.
