CXCR5, C-X-C motif chemokine receptor 5, alias CD185, is a G-protein coupled receptor, a member of the chemokine receptor family, and is expressed predominantly on B-cells, follicular helper T-cells (Tfh), some T-cell subpopulations, and some dendritic cells.The mechanism of action of CXCR5 is through binding to a specific ligand, CXCL13, which activates the receptor and in turn triggers intracellular signaling, such as activation of PI3K, AKT, and ERK/MAPK signaling pathways via Gαi proteins. receptor, which in turn triggers intracellular signaling, such as activation of signaling pathways such as PI3K, AKT, and ERK/MAPK via the Gαi protein, leading to cell migration, proliferation, and differentiation.

In secondary lymphoid organs such as lymph nodes and spleen, the interaction of CXCR5 with its ligand CXCL13 is essential for the formation of B-cell follicles and Tfh-cell compartments, which are critical for the development and maintenance of adaptive immune responses. In addition, CXCR5 also plays a role in certain autoimmune diseases, inflammatory disorders, and tumors, and changes in its expression level may correlate with disease progression. Thus, CXCR5 is a potential target in immunological research and treatment of related diseases.
