FLNA

Filamin A (FLNA), also known as actin-binding protein 280 (ABP-280), is a core cytoskeletal protein that cross-links actin filaments into three-dimensional networks. It regulates cell shape, migration, and mechanotransduction, interacting with Rho GTPase signaling pathways to modulate cytoskeletal dynamics and cell adhesion.
Mutations in FLNA cause X-linked disorders like periventricular nodular heterotopia (PVNH), marked by neuronal migration defects, and skeletal/cardiovascular anomalies such as mitral valve prolapse. No FLNA-targeted drugs are approved yet, but preclinical studies explore Rho kinase inhibitors to mitigate cytoskeletal dysfunctions in related diseases, showing preliminary therapeutic potential.
