The protein encoded by GALT is galactose-1-phosphate uridylyltransferase (EC 2.7.7.12), commonly known as GALT enzyme. It catalyzes the conversion of galactose-1-phosphate and UDP-glucose to UDP-galactose and glucose-1-phosphate, a key step in galactose metabolism that links galactose utilization to glycolysis for energy homeostasis.
Mutations in GALT cause classic galactosemia, an autosomal recessive disorder leading to galactose accumulation, liver damage, cataracts, and neurodevelopmental defects. Current management relies on galactose-restricted diets; no curative drugs exist yet. Preclinical research explores enzyme replacement therapies and CRISPR-Cas9 gene editing to restore GALT activity, targeting potential curative interventions.
