GPSM1 encodes G protein signaling modulator 1, commonly known as AGS3. As a guanine nucleotide dissociation inhibitor (GDI)-like protein, it binds Gαi subunits to sequester them from Gβγ dimers, thereby modulating G protein-coupled receptor (GPCR) signaling cascades. This regulation influences key cellular processes including polarity, spindle orientation, and migration.
Dysregulated GPSM1 is linked to breast and lung carcinomas, where its overexpression correlates with tumor progression and metastasis. It may also contribute to neurological disorders via altering neuronal signaling. Current drug development focuses on targeting its Gαi-binding domains to disrupt aberrant pathways, though such agents remain in preclinical stages.

Immunofluorescence staining of HepG2 cells with CSB-PA801266LA01HU at 1:133, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
