KCNJ3 encodes the G protein-coupled inwardly rectifying potassium channel subunit Kir3.1 (alias GIRK1). It assembles with Kir3.2/Kir3.4 into functional heterotetramers, activated by Gβγ subunits released from G i/o -coupled receptors. This triggers membrane hyperpolarization, dampening neuronal excitability and mediating inhibitory neurotransmission.
Mutations in KCNJ3 are linked to epilepsy, schizophrenia, and cardiac arrhythmias. Drug development focuses on Kir3 channel modulators: positive allosteric modulators show promise for anxiety and pain management, while inhibitors may address certain cardiac conditions. Most candidates remain in preclinical or early clinical phases, with ongoing research into their therapeutic potential.
