The protein encoded by KCNK2 is TWIK-related acid-sensitive potassium channel 1 (TASK-1), a member of the two-pore domain potassium (K2P) channel family. Common aliases include K2P2.1. TASK-1 mediates background potassium currents, regulating membrane potential and cellular excitability, particularly in excitable tissues like the heart and brain. It is sensitive to extracellular pH, with decreased activity under acidic conditions.
TASK-1 functions in pathways regulating respiratory drive and cardiac repolarization. Mutations in KCNK2 are linked to familial atrial fibrillation and idiopathic ventricular fibrillation. Pharmacologically, TASK-1 inhibitors are being developed for treating atrial fibrillation by prolonging action potential duration, with several small-molecule antagonists in preclinical evaluation.
