KDEL endoplasmic reticulum protein retention receptor 2 (KDELR2), also known as ERD2-like protein 2, is a Golgi-localized transmembrane receptor. It mediates retrograde transport of ER-resident proteins bearing the KDEL (Lys-Asp-Glu-Leu) motif from the Golgi back to the ER, ensuring endoplasmic reticulum homeostasis and proper protein folding.
Aberrant KDELR2 expression correlates with breast cancer (overexpression drives tumor aggressiveness) and neurodegenerative disorders linked to ER stress. While no targeted drugs are in clinical trials, preclinical studies explore small-molecule modulators to inhibit its function, aiming to disrupt cancer cell survival or alleviate ER stress-related pathologies.
