KLK7

KLK7, kallikrein-related peptidase 7, also called stratum corneum chymotryptic enzyme, is a secreted serine protease produced mainly by epidermal keratinocytes. It digests corneodesmosomal proteins to control corneocyte shedding and sustain epidermal barrier homeostasis, acting cooperatively with KLK5 within the skin.

KLK7 activity is modulated by endogenous LEKTI. Excessive activation of KLK7 drives barrier impairment, and its elevated levels are observed in Netherton syndrome, atopic dermatitis and psoriasis. Selective small-molecule inhibitors targeting KLK7 are under early pharmacological assessment for inflammatory skin disorders.
