LAMP5

LAMP5, also known as lysosomal associated membrane protein family member 5, is a protein that plays a crucial role in endosomal-lysosomal trafficking and innate immune signaling. It is a member of the LAMP family, which is characterized by its expression in lysosomes and endosomes. LAMP5 functions by regulating the subcellular localization of Toll-like receptors (TLRs), thereby influencing the signaling pathways of interferons and pro-inflammatory cytokines. In the context of leukemia, LAMP5 has been identified as a novel target, particularly in mixed lineage leukemia-rearranged (MLL-r) acute leukemia, where it is overexpressed and directly targeted by the oncogenic MLL-fusion protein. This overexpression leads to the activation of NF-kB signaling and the inhibition of interferon signaling, contributing to the pathogenesis of the disease. The targeting of LAMP5, either through antibody-drug conjugates or other immunotherapeutic approaches, has shown promising results in inhibiting leukemia cell growth and viability.
