LOXL1

LOXL1 (lysyl oxidase-like 1) is a copper-dependent amine oxidase that catalyzes cross-linking of collagens and elastin via oxidative deamination of lysine residues, maintaining extracellular matrix (ECM) integrity. Common aliases include LOX-like protein 1. It regulates ECM remodeling and elastogenesis pathways critical for tissue homeostasis.
Genetic variants in LOXL1 are major risk factors for exfoliation syndrome (XFS) and exfoliation glaucoma (XFG). It also contributes to fibrotic disorders (e.g., liver fibrosis) and cancer metastasis through ECM remodeling. Preclinical research explores LOXL1 inhibitors for fibrosis and modulators for XFG, though no targeted therapies are approved yet.
