MAGEB3

Melanoma Antigen Family B3 (MAGEB3), a cancer-testis antigen (CTA), is encoded by the MAGEB3 gene, with aliases including MAGED and MAGE-B3. Normally restricted to testicular germ cells, it is aberrantly upregulated in melanoma, lung, breast, and prostate cancers, where it promotes cell proliferation via interactions with E3 ubiquitin ligases, regulating protein degradation cascades.
MAGEB3 modulates the p53 signaling pathway, potentially dampening p53’s tumor-suppressive effects. As a CTA, it is a promising immunotherapeutic target: peptide-based vaccines and adoptive T-cell therapies targeting MAGEB3 are under preclinical and early clinical evaluation for MAGEB3-positive malignancies, though no FDA-approved agents exist to date.
