MAP1LC3B

Microtubule-associated protein 1 light chain 3 beta (MAP1LC3B, alias LC3B) is a core autophagy effector. It undergoes lipidation (conjugation to phosphatidylethanolamine via ATG7/ATG3) to anchor to phagophore membranes, driving autophagosome formation. It links nutrient signaling (mTORC1 repression, AMPK activation) to autophagic flux regulation.
Dysregulated MAP1LC3B function associates with neurodegeneration (Alzheimer’s/Parkinson’s, excess autophagosomes) and cancers (colorectal/breast, impaired autophagy). Preclinical agents targeting LC3B lipidation are explored for cancer; rapamycin analogs (mTOR inhibitors) indirectly modulate LC3B clinically, but direct LC3B-targeted drugs remain in early development.
