MGA, recognized as a MAX dimerization protein, is a transcription factor expressed abundantly in tissues like ovaries and testes. This protein, comprising 3042 amino acids, is key in cell fate regulation and in promoting RNA polymerase II-mediated transcription. It upholds the pluripotency of embryonic stem cells and is essential for early embryonic development. MGA is part of the polycomb repressive complex PRC1.6, where it stabilizes the complex and primarily silences genes like Gata4/6 and Sox17 in the primitive endoderm, preventing the differentiation of embryonic stem cells. Therefore, MGA is linked to a range of conditions, from tumors to developmental disorders. Studying MGA deepens our insight into the molecular mechanisms that govern early embryonic cell fate decisions.
