NCOR1 encodes Nuclear Receptor Corepressor 1 (N-CoR), a key transcriptional corepressor. It interacts with nuclear receptors (e.g., RARs, PPARγ) and recruits histone deacetylases (HDACs) to repress target gene expression, modulating pathways like retinoic acid signaling and lipid metabolism to maintain cellular homeostasis.
Dysregulation of NCOR1 is linked to prostate cancer, breast cancer, and metabolic disorders (obesity, type 2 diabetes). Reduced NCOR1 expression in prostate cancer correlates with aggressive phenotypes. Drug development focuses on targeting NCOR1-associated HDAC complexes; some HDAC inhibitors are in clinical trials for cancer, while direct NCOR1 modulators remain in preclinical stages.
