NHERF1

Na+/H+ exchanger regulatory factor 1 (NHERF1), also called ezrin-radixin-moesin-binding phosphoprotein 50 (EBP50), is a scaffold protein with two PDZ domains. It mediates interactions between membrane transporters (e.g., NHE3) and receptor tyrosine kinases (e.g., EGFR), regulating ion balance and signal initiation.
NHERF1 is involved in EGFR/Ras/MAPK and PI3K/Akt pathways; its dysregulation correlates with colorectal, breast, and prostate cancers. Preclinical research focuses on small-molecule inhibitors targeting its PDZ domains to block oncogenic signaling, though no candidates have advanced to clinical trials.
