NLRP10, also known as NOD8, PAN5, PYNOD, NALP10, and CLR11.1, contains the pyrin domain 10 of the NLR family. As part of a signaling-induced multimeric complex (inflammatory bodies), NLRP10 activates pro-inflammatory cysteine proteases caspase-1 and caspase-5. Additionally, NLRP10 is considered a multifunctional negative regulator of inflammation and apoptosis. In the innate immune system, NLRP10 plays a regulatory role in defense responses against other organisms, positive regulation of adaptive immune responses through somatic recombination of immunoreceptors based on immunoglobulin superfamily domains, and positive regulation of cytokine production. NLRP10 is associated with various diseases, including Petrositis and neurodevelopmental disorders with involuntary movements.

IHC image of CSB-PA773044LA01HU diluted at 1:50 and staining in paraffin-embedded human prostate cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB. Secondary antibody only control: uses 1% BSA instead of primary antibody
Immunofluorescence staining of Hela cell with CSB-PA773044LA01HU at 1:20, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and and permeated by 0.2% TritonX-100 for 15 min. Then 10% normal goat serum to block non-specific protein-protein interactions . The cells were then incubated with the antibody overnight at 4℃. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
Immunofluorescence staining of Hela cell with 5% goat serum, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
