PEMT

Phosphatidylethanolamine N-methyltransferase (PEMT), also referred to as PE N-methyltransferase, is a core enzyme in hepatic phosphatidylcholine (PC) biosynthesis. It mediates sequential methylation of phosphatidylethanolamine (PE) using S-adenosylmethionine (SAM) as the methyl donor, generating PC—an essential component of cell membranes and lipoproteins. This process sustains lipid homeostasis and supports liver function by regulating lipoprotein secretion.
Dysfunctional PEMT is associated with non-alcoholic fatty liver disease (NAFLD): reduced activity impairs PC synthesis, driving hepatic lipid accumulation and steatosis. It may also link to cardiovascular risks via altered lipoprotein metabolism. Preclinical research explores small-molecule PEMT activators for NAFLD, with promising data on mitigating liver fat, though clinical trials remain early-stage.
