PIM3

Proviral Integration Site 3 (PIM3), a serine/threonine kinase also called PIM-3, is a member of the PIM family. It modulates cell proliferation, survival, and metabolism by phosphorylating substrates like BAD (suppressing apoptosis), p21 (driving cell cycle), and AKT (amplifying signaling), exerting pro-tumorigenic effects via anti-apoptotic and growth-promoting actions.
PIM3 interacts with PI3K/AKT, JAK/STAT, and MAPK pathways. Its overexpression links to poor outcomes in digestive cancers (liver, pancreatic, colorectal). Pan-PIM inhibitors (e.g., SGI-1776) show preclinical anti-tumor activity, while selective PIM3 inhibitors targeting metabolic reprogramming remain in early development, promising precision therapeutic potential.
