PLP1

Proteolipid protein 1 (PLP1), also known as DM20 (its alternatively spliced isoform), is the most abundant integral membrane protein in central nervous system (CNS) myelin, accounting for ~50% of total myelin proteins. It plays a critical role in maintaining myelin structural integrity and stability, facilitating efficient axonal signal conduction by forming compact myelin sheaths around axons.
Mutations in PLP1 cause Pelizaeus-Merzbacher disease (PMD), an X-linked recessive disorder characterized by impaired myelinogenesis and progressive neurological deficits. Current therapeutic efforts focus on gene therapy (e.g., AAV-mediated PLP1 expression modulation) and small molecules targeting myelin repair pathways, though no approved treatments exist yet.
