PPTC7

The PPTC7 gene encodes the protein phosphatase Mg²⁺/Mn²⁺ dependent 1G (PPM1G), a serine/threonine phosphatase also known by aliases like PPM1G. It regulates cellular responses to DNA damage by dephosphorylating key substrates such as p53 at Ser392, modulating p53 stability and its downstream functions in cell cycle arrest and apoptosis.
Dysregulation of PPTC7 is linked to breast and lung cancers, where its overexpression may suppress p53-mediated tumor suppression. While no targeted drugs are approved, preclinical studies explore small-molecule inhibitors of its phosphatase domain to reactivate p53, showing promising in vitro efficacy against p53-deficient tumor cell lines.
