PREP, also known as prolyl endopeptidase, is a serine protease widely distributed in the brain, liver and immune cells. It specifically cleaves peptide bonds at proline residues, regulating the metabolism of neuropeptides and hormones to maintain neural and cellular homeostasis.
Dysregulated PREP activity is associated with neurodegenerative diseases such as Alzheimer’s and Parkinson’s, as well as certain cancers and inflammatory disorders. Multiple small-molecule PREP inhibitors are under preclinical and early clinical development, showing potential for treating neurodegenerative conditions.
