SLC38A9, formally solute carrier family 38 member 9 (alias SNAT9), is a lysosomal membrane transporter mediating arginine and glutamine transport. It acts as a critical amino acid sensor for the mTORC1 pathway: upon binding arginine, it interacts with Rag GTPases and Ragulator, activating mTORC1 to regulate cell growth, proliferation, and autophagy.
Dysregulation of SLC38A9 links to mTORC1-related disorders like renal cell carcinoma and metabolic syndromes. Preclinical studies explore small-molecule inhibitors targeting SLC38A9 to modulate mTORC1 signaling, offering potential cancer and metabolic disease therapies, though no drugs are yet clinically approved.
