SLFN11

SLFN11, known as Schlafen 11, is an interferon-induced antiviral restriction factor with tRNA endonuclease and DNA-binding functions. It is recruited to stalled replication forks during replication stress and may irreversibly trigger replication blockage and cell death.The gene for SLFN11 is involved in cell fate determination of cells following chemotherapy and encodes the N-terminal ribonuclease (RNase) structural domain and the C-terminal deconjugase/ATPase structural domain. The contribution of these structural domains to the response to chemotherapy remains controversial.The dynamic expression of SLFN11 in circulating tumour cells serves as a liquid biomarker for small-cell lung cancer, and its expression levels are lower in patients treated with platinum-based therapies and correlate with clinical response. SLFN11 is a promising biomarker in predicting response to treatment for multiple drug-induced DNA damage, including platinum-based drugs and PARP inhibitors.
