SLIT3

SLIT3 encodes Slit Guidance Ligand 3, a secreted glycoprotein of the SLIT family. Common aliases include FLJ20176 and Drosophila slit homolog 3. It binds ROBO receptors to activate the SLIT-ROBO pathway, regulating cytoskeletal dynamics via Rho GTPases to mediate repulsive cell guidance—critical for vascular development and lung branching morphogenesis.
Mutations in SLIT3 are linked to congenital diaphragmatic hernia (CDH) due to impaired diaphragm formation. Reduced SLIT3 expression correlates with enhanced metastasis in colorectal cancer, as it inhibits cell migration. Preclinical studies explore SLIT3-based therapies for metastatic cancers and vascular disorders, though no approved drugs targeting SLIT3 exist yet.
