TBL2, also known as transmembrane protein β-like 2, has alternative names including WBSCR13 and WS-betaTRP. TBL2 is an endoplasmic reticulum (ER) localized type I transmembrane protein with a WD40 repeat sequence and a coiled helix domain. It plays a role in regulating the ER stress signaling pathway. TBL2 specifically binds to the phosphorylated form of PERK (PKR-like ER resident kinase), interacting with PERK through its proximal N-terminal region and binding to eIF2α via the WD40 domain. Knockdown of TBL2 leads to impaired induction of ATF4 (activating transcription factor 4) under ER stress or malnutrition conditions, such as glucose and oxygen deprivation, reducing cellular resistance to stress, similar to cells with PERK knockdown. As a potential regulator of the PERK pathway, TBL2 is crucial for cell survival under ER stress and malnutrition conditions. The TBL2 gene is absent in Williams-Beuren syndrome, a developmental disorder caused by multiple gene deletions at 7q11.23.
