TCF12, transcription factor 12, alias HEB, p64, CRS3, HH26, HTF4, TCF-12, bHLHb20, HsT17266, etc. TCF12 is a member of the basic helix-loop-helix (bHLH) family of E proteins that recognizes a specific binding site (E-box), "CANNTG". It is expressed in a variety of tissues, including skeletal muscle, thymus, B cells, and T cells, and may be involved in the regulation of lineage-specific gene expression through the formation of heterodimers with other bHLH E proteins.TCF12 has a wide range of biological significance, and it is involved in the regulation of a wide range of cellular developmental and differentiation processes. For example, TCF12 is highly expressed in hematopoietic stem cells and plays an important role in regulating their function and differentiation to lymphocyte precursors. In addition, TCF12 is associated with cranial growth and motor learning, and its loss-of-function mutations can lead to premature closure of the cranial sutures and neurodevelopmental delays. In osteoarthritis, TCF12 regulates the TGF-β/Smad2/3 signaling pathway by targeting CXCR4, accelerating disease progression. Thus, TCF12 plays a key role in maintaining normal physiological function and disease development.
