UDP-glucose 6-dehydrogenase (UGDH), also known as UGD or UDPGD, is a key enzyme in the glucuronate pathway. It catalyzes NAD+-dependent oxidation of UDP-glucose to UDP-glucuronic acid—a precursor for glycosaminoglycans (GAGs) like hyaluronan and chondroitin sulfate, critical components of the extracellular matrix (ECM).
UGDH activity is linked to ECM remodeling, regulated by pathways such as TGF-β. Dysregulation causes fibrotic disorders (liver/renal fibrosis) via excessive GAG accumulation; its overexpression in cancers drives metastasis. Small-molecule inhibitors targeting UGDH are under investigation as potential therapeutics for fibrosis and malignancies.
