ZPR1

ZPR1 (zinc finger protein ZPR1 homolog), also known as ZNF259, is a conserved zinc finger-containing protein critical for ribosome biogenesis and cell cycle progression. It functions as a downstream target of the Ras/ERK pathway: ERK-mediated phosphorylation drives ZPR1 nuclear translocation, where it interacts with ribosomal subunits to support ribosome assembly, thus promoting cell proliferation and survival.
Abnormal ZPR1 expression is associated with breast, lung, and colorectal cancers—its overexpression correlates with tumor growth and poor prognosis. Preclinical studies explore targeting ZPR1’s interaction with ribosomal proteins or its phosphorylation by ERK to inhibit tumorigenesis, though no specific drugs have advanced to clinical trials yet.
