Perfringolysin O (PFO) is a toxin produced by Clostridium perfringens bacteria. Its structure consists of a large single-subunit protein of approximately 497 amino acids, containing multiple β-folds and α-helical structures. PFO primarily binds to cholesterol in cell membranes, forming polymers and creating permeable pores, resulting in the disruption and dissolution of the cell membrane. This pore-forming action compromises cell integrity, leading to lysis and cell death. By binding predominantly to cholesterol and initiating the dissolution of cell membranes, PFO disrupts the ion balance inside and outside cells, affecting cell viability and function. PFO and its variants serve as tools in biological research for cell membrane lysis, aiding the investigation of cell membrane permeability and the effects of membrane damage. In cases of Clostridium perfringens infection, PFO is considered a significant virulence factor, contributing to cell lysis and tissue damage.
