ACSL4

ACSL4, located on Xq22.3~23, is central to diverse biological functions. This enzyme is particularly abundant in tissues like the adrenal glands and appendix. ACSL4 specializes in converting free long-chain fatty acids into acyl-CoA esters, significantly impacting lipid synthesis and the breakdown of fatty acids, with a particular preference for arachidonic acid. ACSL4's influence extends to the modulation of lipid metabolism, inflammation, insulin release, muscle protein turnover, and cell death pathways like ferroptosis. Dysregulation of ACSL4 is linked to an array of pathologies, such as non-alcoholic fatty liver disease, various cancers, ischemic stroke, and acute kidney injury, positioning ACSL4 as a promising target for therapeutic intervention across several disease states.
