ADAM10 (A Disintegrin And Metalloproteinase 10), also known as Kuzbanian homolog or CD156c, is a transmembrane zinc-dependent protease critical for regulated proteolysis of cell surface proteins. It mediates ectodomain shedding of substrates including Notch receptors, amyloid precursor protein (APP), and adhesion molecules, thereby modulating cell signaling, adhesion, and migration. ADAM10 is indispensable for Notch signaling activation during embryonic development and tissue homeostasis. Its sheddase activity also regulates growth factor release (e.g., EGFR ligands) and immune responses through cleavage of cytokines like TNF-α. Overlay Peak curve showing Hela cells stained with CSB-RA001273MA1HU (red line) at 1:400. The cells were fixed in 4% formaldehyde (15min) and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum was Incubated to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 45 min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-Mouse IgG(H+L) at 1/200 dilution for 35 min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1µg/1*106cells) used under the same conditions. Acquisition of >10, 000 events was performed.

Immunofluorescence staining of Hela cells with CSB-PA001273LA01HU at 1:133, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
