ALPK1, also known as α-kinase 1, and by its aliases LAK and ROSAH, is an atypical serine/threonine protein kinase that lacks sequence homology with conventional protein kinases. ALPK1 is expressed in a variety of tissues, including bone marrow and skin.
The mechanism of action of ALPK1 involves the recognition and specific binding to ADP-heptose, a precursor sugar molecule in the synthesis of bacterial lipopolysaccharide (LPS), which subsequently activates the NF-κB pathway to mediate innate immune inflammatory responses.
As a pattern recognition receptor (PRR), ALPK1 plays a crucial role in the innate immune response, particularly in the recognition of pathogen-associated molecular patterns (PAMPs) and the activation of host immune responses.
