ATP5F1A encodes the ATP synthase F1 subunit alpha (ATP5A1), a core component of the mitochondrial oxidative phosphorylation (OXPHOS) system. Common aliases include F1-ATPase alpha chain. It forms the catalytic core of the F1 complex with beta subunits, catalyzing ATP synthesis from ADP and inorganic phosphate by harnessing the proton gradient across the inner mitochondrial membrane.
Mutations in ATP5F1A cause mitochondrial complex V deficiency, leading to encephalomyopathy, lactic acidosis, and cardiomyopathy. Dysregulated expression is linked to cancers (breast, lung) via enhanced tumor energy metabolism. Preclinical efforts focus on gene therapy and mitochondrial function modulators; oligomycin, an ATP synthase inhibitor, is a research tool but not yet a therapeutic for ATP5F1A-related disorders.
