CALCR, found on human chromosome 19, is a protein that resides in the endoplasmic reticulum, nucleus, and cell membrane, and is integral to the calcitonin signaling system. This receptor activates the G protein-coupled receptor signaling pathway upon binding to calcitonin, which regulates blood calcium levels, curbs bone resorption, and enhances renal calcium excretion. The presence of CALCR in the brain hints at its potential roles in the central nervous system, including the modulation of pituitary hormone secretion, appetite suppression, gastric acid release, and it may be involved in pain regulation or autism pathogenesis. Crucial for bone health, CALCR is a significant player in bone disease, with mutations linked to osteoporosis and aberrant expression observed in various tumors.
