The protein encoded by CYP21A2 is steroid 21-hydroxylase (EC 1.14.99.10), commonly known as P450c21. As a key cytochrome P450 enzyme in the adrenal cortex, it catalyzes the 21-hydroxylation of progesterone and 17α-hydroxyprogesterone to 11-deoxycorticosterone and 11-deoxycortisol—critical steps for glucocorticoid and mineralocorticoid biosynthesis.
Mutations in CYP21A2 cause congenital adrenal hyperplasia (CAH), the most common adrenal disorder in children, marked by cortisol deficiency and excess androgens. Current treatments include glucocorticoid replacement (e.g., hydrocortisone), while emerging therapies like CYP11B2 inhibitors and gene editing approaches are under investigation to address unmet clinical needs.

IHC image of CSB-PA006400LA01HU diluted at 1:100 and staining in paraffin-embedded human adrenal gland tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
Immunofluorescence staining of HepG2 with CSB-PA006400LA01HU at 1:33, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
