DNA2 is an enzyme with both nuclease and helicase domains, playing a pivotal role in DNA replication and repair. It removes RNA primers during Okazaki fragment maturation and performs end resection during double-stranded DNA break repair. Additionally, DNA2 stabilizes and restarts stalled replication forks with chicken-foot structures. In humans, DNA2 is primarily located in mitochondria, participating in mitochondrial DNA replication and repair processes, including the removal of RNA primers and enhancing the efficiency of poly(dA) synthesis. Although we have a preliminary understanding of the structure and function of DNA2, its specific role in signaling pathways and interactions with other molecules remain to be further explored. Such studies will not only deepen our understanding of DNA replication and repair mechanisms but also provide crucial insights for the development of novel therapeutic strategies.
