FPGS

FPGS, folylpolyglutamate synthetase, alias Folylpoly-gamma-glutamatesynthetase, is an enzyme expressed in human cells that is essential for folate homeostasis and proliferating cell survival.
The mechanism of action of FPGS is to establish and maintain folatesynthetase in the cytosol and in the mitochondria by catalyzing the ATP-dependent addition of glutamate fractions to folate and folate derivatives in the lysosomes and mitochondria to establish and maintain folyl polyglutamate concentrations. The activity of this enzyme is critical for the intracellular metabolism process of methotrexate (MTX)-like drugs, which catalyzes the formation of MTX polyglutamate, the active form of MTX that exerts antitumor effects.
Abnormal expression or reduced activity of FPGS may lead to MTX resistance, and the expression and activity of FPGS is an important factor affecting drug sensitivity in cancer therapy.
