GNA13

Guanine nucleotide-binding protein G(13) subunit alpha (GNA13, alias Gα13) is a heterotrimeric G protein alpha subunit. It transduces G protein-coupled receptor (GPCR) signals via GDP-GTP exchange, activating Rho guanine nucleotide exchange factors (RhoGEFs) like ARHGEF1/11 to trigger RhoA GTPase activation—regulating cytoskeletal rearrangement, cell adhesion, and migration.
Dysregulated GNA13 links to vascular anomalies (e.g., venous malformations from gain-of-function mutations) and cancers (promoting invasion/metastasis in colorectal and breast tumors). Drug development targets the Gα13-RhoA pathway; preclinical candidates include small molecules inhibiting Gα13-RhoGEF interactions or Rho-associated kinase (ROCK), though no FDA-approved therapies exist for GNA13-related disorders yet.
