HLA-G

HLA-G, also known as HLA class Ib molecule, is a non-classical MHC protein expressed on various immune cells and tumor cells. It interacts with inhibitory receptors like ILT2, ILT4, and KIR2DL4, leading to immune suppression by inhibiting T cell proliferation, NK cell cytotoxicity, and B cell activation. HLA-G is involved in immune tolerance, cancer immune evasion, and autoimmune diseases. Its expression is regulated by epigenetic modifications, transcription factors, and post-translational modifications. HLA-G is a potential target for cancer immunotherapy and autoimmune disease treatment. Research on HLA-G-based therapies is ongoing, with monoclonal antibodies and immune checkpoint inhibitors being investigated.

Overlay Peak curve showing U937 cells stained with CSB-RA117800A0HU (red line) at 1:50. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1µg/1*10⁶cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1µg/1*10⁶cells) used under the same conditions. Acquisition of >10,000 events was performed.
