IMPDH2

IMPDH2 encodes Inosine Monophosphate Dehydrogenase 2, a rate-limiting enzyme in de novo guanine nucleotide biosynthesis. Its core function is catalyzing the oxidation of inosine monophosphate (IMP) to xanthosine monophosphate (XMP) using NAD+ as a cofactor—this step is critical for generating GTP and GDP, which support DNA/RNA synthesis and cell proliferation. Common aliases include IMD2 and IMPDH type II. The protein is regulated by pathways like PI3K/Akt and mTOR, which modulate its activity to meet nucleotide demands in rapidly dividing cells.
Dysregulation of IMPDH2 is associated with multiple diseases: overexpression occurs in cancers (leukemia, colorectal cancer) due to tumor cells’ high nucleotide needs, and it contributes to autoimmune disorders (rheumatoid arthritis) via excessive immune cell proliferation. Mycophenolic acid (MPA), an IMPDH inhibitor, is an FDA-approved immunosuppressant for organ transplantation. Novel isoform-selective IMPDH2 inhibitors are in preclinical/clinical development for cancer, targeting its preferential expression in proliferating malignant cells to reduce off-target effects.

Overlay Peak curve showing jurkat cells stained with CSB-RA210986A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*10⁶cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*10⁶cells) used under the same conditions. Acquisition of >10,000 events was performed.
