IRAK4

IRAK4, interleukin-1 receptor-associated kinase 4, alias IL-1 receptor-associated kinase 4, is a key protein in Toll-like receptors (TLRs) and IL-1 receptor-mediated inflammation.IRAK4 is encoded by its own genes in human cells, and its mechanism of action is mainly to induce inflammatory factor expression by participating in the Toll-like receptor signaling pathway and activating downstream NF-κB and AP-1 transcription factors, thereby inducing the expression of inflammatory factors, cytokines, and chemokines.

IRAK4 is biologically very important, and it is not only involved in the positive regulation of inflammatory responses, but also extensively involved in the networked regulation of intracellular responses.Abnormal function of IRAK4 has been associated with the development of a variety of inflammatory diseases, including rheumatoid arthritis, atopic dermatitis, pyogenic sweating inflammation, and so on. In addition, overexpression of IRAK4 enhances LPS-induced NADPH oxidase activity and induces downstream signaling of p38 MAPK, further expanding the biological functions of IRAK4 . Thus, IRAK4 is an important target in anti-inflammatory and tumor research, and small molecule inhibitors and protein degraders against IRAK4 are being developed for the treatment of related diseases.
