KLHDC3 encodes Kelch domain-containing protein 3, a substrate adaptor for Cullin-RING E3 ubiquitin ligases. It mediates ubiquitination and proteasomal degradation of target proteins, regulating cellular proliferation and differentiation. A common alias is KIAA1468.
Dysregulation of KLHDC3 is linked to lung and breast cancers; its overexpression in tumors correlates with aggressive growth and poor prognosis, likely via modulating oncogenic signaling pathways. While no KLHDC3-targeted drugs are clinically available, preclinical studies explore inhibiting its ligase activity as a potential therapeutic strategy.
