LZTR1 encodes the leucine zipper-like transcriptional regulator 1, also known as KIA0317. As an adaptor subunit of CUL3-RING E3 ubiquitin ligase complexes, it mediates ubiquitination and proteasomal degradation of RAS family proteins (e.g., RIT1, HRAS), thereby negatively regulating the RAS-MAPK signaling pathway that governs cell proliferation and differentiation.
Mutations in LZTR1 are associated with neurofibromatosis type 2 (NF2), schwannomatosis, and certain Noonan syndrome-spectrum disorders. Current drug development focuses on targeting downstream RAS-MAPK cascade components (e.g., MEK inhibitors) for NF2-related tumors, while direct modulators of LZTR1-CUL3 activity remain in preclinical research stages.
